Fraction I became known as PRPP synthetase, Fraction II as hypoxanthineguanine 5'-phosphoribosyl transferase [75,79]. The latter enzyme also utilizes guanine and the fraudulent purine, 6-mercaptopurine [80], as substrates. A second phosphoribosyl transferase catalyzes the reaction of PRPP with adenine [75, 79] and 5-amino-4-imidazole carboxamide [81] to their corresponding ribotides. The reactions catalyzed by the purine phosphoribosyl transferases have been referred to commonly as the salvage pathway of nucleotide synthesis.

I particularly remember conversations with Rollin Hotchkiss about his work on DNA as a transforming factor for pneumococcal bacteria as a possible subject for a new research project. As interesting as this research was from the point of view of the geneticist, I did not see how a biochemist could contribute to such a difficult problem at that time. On the other hand I recalled work done at the University of Pennsylvania by my colleague, Seymour S. Cohen who, with Wyatt [157], had identified 5-hydroxymethylcytosine as replacing cytosine in the DNA formed in E.

The former may also be grown in a methionine-free medium to which vitamin B 12 is supplied. Thus, an extract of either mutant by itself was incapable of synthesizing methionine from a reaction mixture containing ATP, 5,10-methylene tetrahydrofolate, homocysteine and a reducing source, but when combined they could [138-142]. The problem then was to develop an assay so that either enzyme could be isolated and purified. By using the strategy employed so frequently in our studies on purine biosynthesis, one enzyme was purified in the presence of an excess of the other and vice versa without knowledge of the characteristics or intermediates of the reaction.
